摘要
目的研究RhoA和Rock-Ⅱ在Lewis大鼠实验性自身免疫性脑脊髓炎(EAE)模型中的表达,探索其在EAE模型中的作用。方法把36只Lewis大鼠随机分为两组:A组(空白对照组),B组(EAE模型组)。以豚鼠全脊髓为抗原混合福氏完全佐剂(CFA),诱导Lewis大鼠EAE模型,观察实验动物神经功能损害的情况;采用免疫组织化学染色法观察实验动物各组脑、脊髓组织中RhoA、Rock-Ⅱ的表达。结果成功建立Lewis大鼠EAE模型,RhoA和Rock-Ⅱ在EAE模型组的表达较空白对照组明显增加,差异有统计学意义(P<0.01)。结论 RhoA和Rock-Ⅱ在EAE模型中过量表达,可能在EAE模型的发病过程中起关键作用。
Objective To investigate the expression of RhoA and Rock-Ⅱin brain and spinal cords,and to explore its effect in the experimental autoimmune encephalomyelitis(EAE).Methods 36 Lewis rats were divided into two groups:Lewis rats control(A),and EAE group(B).The EAE model was established in Lewis rats by immunization with complete Freunds adjuvant(CFA)and Guinea pig spinal cord homogenate(GPSCH).The experimental autoimmune encephalomyelitis(EAE) was determined by clinical scores.The expression levels of RhoA and Rock-Ⅱin brain and spinal cords were determined by histochemistry.Results The EAE animal model was established successfully.The results showed that the expression levels of RhoA and Rock-Ⅱin brai and spinal cords were significantly increased in the EAE model compared with control group(P0.01).Conclusion The overexpression of RhoA and Rock-II in the EAE model may play an important role on the effector phase of EAE.
