摘要 目的:了解肝损伤大鼠在自体动员和移植骨髓干细胞后肝功能、肝病理变化情况,为临床应用提供实验依据.方法:将大鼠随机分为正常对照组和造模组.造模组采用腹腔注射四氯化碳的方法构建大鼠肝损伤模型,然后随机分为粒细胞集落刺激因子(G-CSF)动员组、干细胞移植组和模型对照组,G-CSF动员组采用注射G-CSF、干细胞移植组经门静脉输注骨髓间充质干细胞.比较两种治疗方法治疗肝损伤的效果.结果:治疗3周后,干细胞移植组的肝功能、肝病理变化较模型对照组明显好转,但G-CSF动员组效果不明显且两者均未使肝功能等恢复到正常.结论:骨髓间充质干细胞移植治疗大鼠肝损伤,疗效好于G-CSF动员. Objective: To understand liver function and liver pathological changes after mobilization and transplantation of autologous bone marrow stem cells in rats with liver injury, and to provide experimental basis for clinical application. Methods: The rats were randomly divided into a normal control group and a model group. Using intraperitoneal injection of carbon tetrachloride, the liver injury model was induced, and the model group were randomly divided into a G-CSF mobilization group, a stem cell transplantation group and a model control group. G-CSF mobilization group were treated with injection of G-CSF, stem cell transplantation group through the portal vein infusion of bone marrow-derived mesenchymal stem cells. Liver function and liver pathological changes were detected, and the two treatment methods were compared to restore the effect of liver damage. Results. After 3 weeks, liver function and pathological changes in the stem cell transplantation group were significantly improved compared with the model group, and G-CSF mobilization group had no significant recoverier of liver functions compared with model control group, both of which did not return to normal. Conclusion: Transplantation of bone marrow mesenchymal stem ceils for the treatment of rat liver injury is more effective than G-CSF mobilization.
