摘要 目的:建立大鼠小脑外形和小脑深部核团三维可视化模型.方法:用冷冻切片机对小脑组织连续切片、照相,并依次编号,裱片后进行尼氏染色,再对染色后的切片进行拍照,从而获取两份相对应的原始图像数据库.应用Photoshop 9.0对数据库中的数据进行半自动配准及人工分割,应用Amira 4.1.1软件对分割后的数据库进行三维重建.结果:获得大鼠小脑外形及小脑深部核团微细结构信息的数据库.运用Amira 4.1.1应用软件可分别对小脑外形及小脑核团进行三维显示,成功地定位了小脑深部核团在小脑中的空间位置形态以及与小脑周围组织的毗邻关系.结论:采用数字人图像处理技术配合传统的染色技术,可以获取理想的三维图像信息,更为真实,层次感更强.此法适用于小脑外形及小脑深部核团等组织精细结构的三维重建. Objective: To establish a detailed digitalized dataset for 3D reconstruction of the rat cerebellum and its deep cere- bellar nuclei. Methods : The rat cerebellum was sectioned serially. Then the sections were stained with Nissel's method, and photographed with the digital camera. The original images dataset was received, aligned semi-automatically and segmented manually using Photoshop 9.0, and the segmented structures were reconstructed using Amira 4.1.1. Results: The original dataset of the detail morphological structures of rat cerebellum and the deep cerebellar nuclei was made up. They were chosen to make 3D reconstruction with Amira 4.1.1 software. In 3D model, the position and relationship of the deep cerebellar nuclei which distinguished from the surroundings structures were observed clearly. Conclusion: 3D digital model of cerebellum was successfully obtained combined with digital human techniques and histoehemical staining, with full information, real structure and strong laminar. This method is proper for 3D reconstruction of detail microstructures.
