摘要
目的:了解半乳糖凝集素-3(galectin-3)在早孕期绒毛与蜕膜中的表达,为进一步探讨稽留流产发病机制提供理论依据。方法:分别获取手术流产组(n=20)、药物流产组(n=20)及稽留流产组(n=20)患者的流产绒毛及蜕膜,用免疫组织化学法及实时荧光相对定量PCR法检测各组绒毛及蜕膜组织中半乳糖凝集素-3的表达。结果:免疫组织化学显示稽留流产组绒毛半乳糖凝集素-3强阳性表达率(30.00%)明显低于手术流产组(80.00%)(P<0.01)。RT-PCR结果进一步证实稽留流产组绒毛半乳糖凝集素-3 mRNA的相对表达量(2-△△Ct=0.31±0.70)明显低于手术流产组(2-△△Ct=1.00±0.00)(P<0.01),药物流产组蜕膜半乳糖凝集素-3 mRNA的相对表达量(2-△△Ct=0.74±2.20)明显低于手术流产组(2-△△Ct=1.00±0.00)(P<0.01)。结论:半乳糖凝集素-3与妊娠早期绒毛及蜕膜发育有关,其表达降低可能促进了稽留流产的发生。
Objective:To detect the expression of galectin-3 in villus and decidua of early pregnancy,and explore the pathogenesis of missed abortion.Methods:Sixty cases of villus and decidua were divided into 3 groups:surgical abortion group(n=20),medical abortion group(n=20) and missed abortion group(n=20).The expression of galectin-3 in villus and decidua of each group was measured by methods of immunohistochemis-try and real-time fluorescence relative quantitative PCR.Results:Immunohistochemistry showed that galectin-3 strongly-positive expression in villus of missed abortion group(30.00%) was significantly lower than that of surgical abortion group(80.00%)(P0.01).Real-time fluorescence relative quantitative PCR showed that galectin-3 mRNA relative expression in villus of missed abortion group(2-△△Ct =0.31±0.70) was significantly lower than that of surgical abortion group(2-△△Ct =1.00±0.00).Galectin-3 mRNA relative expression in decidua of medical abortion group(2-△△Ct =0.74 ± 2.20) was significantly lower than that of surgical abortion group(2-△△Ct =1.00±0.00)(P0.01).Conclusion:Galectin-3 is related to the development of villus and decidua during early pregnancy.The decreased expression of galectin-3 may promote the occurrence of missed abortion.
