乌骨藤制剂对人肝癌细胞株VEGF和bFGF表达的影响 被引量:15

摘要 目的探讨乌骨藤制剂对人肝癌细胞株VEGF和bFGF表达的影响及其抗肿瘤的作用机制。方法采用ELISA法和细胞免疫化学SP染色法观察不同浓度的乌骨藤制剂作用人肝癌Bel-7402细胞24h、48h后VEGF和bFGF的表达。结果不同浓度的乌骨藤制剂(10mg/ml、20mg/ml、40mg/ml)作用Bel-7402细胞后,VEGF和bFGF表达均随药物浓度的增加逐渐减少,与阴性对照组相比有显著性差异。结论乌骨藤制剂能够明显抑制人肝癌Bel-7402细胞VEGF和bFGF的表达,提示乌骨藤制剂抗肝癌的作用机制可能与抑制肿瘤血管生成密切相关。 Objective To study effects of traditional Chinese medicine glaucescent fissistigma root on hepatocarcinoma cell line,and to clarify the anti-tumor mechanism according to the expressions of vascular endothelial growth factor(VEGF)and basic fibroblast growth factor(bFGF).Methods The inhibiting rate of glaucescent fissistigma root on tumor cell was detected by MTT deoxidized method in vitro.Hepatocarcinoma cell line Bel-7402 was affected by different concentrations of glaucescent fissistigma root.Dosage-effectiveness atlas was graphed on the basis of optical density value.The expressions of VEGF and bFGF in different time and different concentrations of glaucescent fissistigma root was detected by ELISA and immunocytochemical methods respectively.Results Compared to negative control group,different concentrations of glaucescent fissistigma root(10,20,40mg/ml)could inhibit the expressions of VEGF and bFGF(P0.01),which was in dose-dependent manner.Conclusion Glaucescent fissistigma root had strong anti-tumor effects in human hepatocarcinoma cell Bel-7402 in vitro.It can inhibit VEGF and bFGF in the expression of angiogenesis.The mechanism was probably related to the inhibition of angiogenic factors to block tumor angiogenesis.
