摘要
目的探讨肝癌衍生生长因子(Hepatoma-derived growth factor,HDGF)在乳腺癌组织中的表达及其意义。方法应用逆转录聚合酶链反应(Reverse-transcription polymerase chain reaction,RT-PCR)方法检测24例乳腺癌及其配对正常乳腺组织中HDGF mRNA的表达;应用免疫组织化学SP法检测75例乳腺癌及其配对正常乳腺组织中HDGF蛋白的表达水平。结果乳腺癌组织和正常乳腺组织中HDGF基因的表达强度分别是看家基因β-actin的(2.00±0.85)和(0.67±0.37)倍,正常乳腺组织中HDGF mRNA表达明显低于乳腺癌组织中的表达(P<0.001);HDGF蛋白在乳腺癌组织的表达水平明显高于正常乳腺组织(P<0.001);乳腺癌临床分期越高,HDGF的表达水平越高;淋巴结转移阳性的乳腺癌病例中HDGF的表达水平明显高于淋巴结转移阴性组,差异具有统计学意义(P>0.05)。结论 HDGF高表达可能促进了乳腺癌的发病过程,并与乳腺癌淋巴结转移相关。
Aim To investigate the relationship between the expression of hepatoma-derived growth factor(HDGF)and its clinical significance in human breast cancer tissues.Methods The expression of HDGF mRNA was detected in 24 human fresh breast cancer cases and the corresponding adjacent normal mammary tissues by reverse transcriptase-polymerase chain reaction(RT-PCR),and immunohistochemistry was used to detect HDGF protein expression in 75 human breast cancer cases and the corresponding adjacent normal mammary tissues Results The expression of HDGF gene in cancer tissues and benign mammary tissues was 2.00±0.85 and 0.67±0.37 than that of housekeeping gene.The expression of HDGF mRNA in breast cancer was significant higher than that of normal mammary tissues(P0.001).Compared with primary breast cancers,a significant reduction in expression of HDGF protein was noted in benign mammary lesions(P0.001).The expression level of HDGF in high-stage breast cancers was higher than that of low-stage breast cancers.Moreover,the expression of HDGF protein was markedly increased in breast cancers with lymph node metastasis compared with those without metastasis(P0.05).Conclusion Increased expression of HDGF may play a role in the pathogenesis and metastasis of breast cancer.
