不同商品等级枸杞子的抗氧化活性比较及其生物效价检测 被引量:11

摘要 目的:针对枸杞子的抗氧化作用,尝试建立基于清除有机自由基1,1-二苯基苦基苯肼(DPPH)的枸杞子质量控制方法。方法:通过正交试验,优选枸杞子样品提取方法;通过单因素考察确定枸杞子与DPPH溶液的最佳反应条件。结果:固液比1∶10,50%乙醇超声提取20 min时自由基清除率最高;DPPH浓度0.05 mg.mL-1,样品浓度100 mg.mL-1,室温反应90 min为最适反应条件。不同商品等级的枸杞子自由基清除率不同。结论:文中建立的抗氧化生物效价检测法,以枸杞子对DPPH系统自由基清除率为评价指标,可以客观反映枸杞子抗氧化活性的差异,适用于枸杞子药材质量控制,对其他具有抗氧化作用中药的质量控制以及高通量活性筛选也具有一定的参考价值。 Objective: To establish a quality control method for fructus lycii based on its antioxidation scavenging 1,1-diphenyl-2-picrylhydrazyl(DPPH).Methods: Through orthogonal experiments,the extraction technology of fructus lycii was optimized,and the best reaction conditions were investigated with single-factor analysis.Results: The conditions included solid-liquid ratio 1∶10,and solvent of 50% ethanol,and extraction method of ultrasounding for 20 min were carried out for extracting fructus lycii samples.Then,the antioxidative activity of fructus lycii was detected.The optimal reaction system,0.05 mg·mL^-1 DPPH plus 100 mg·mL^-1 sample solution,was reacted for 90 min at room temperature.fructus lycii of different commercial grades exerted various scavenge rates of DPPH.The biological activity was higher in samples from high grade fructus lycii.Conclusion: The established bioassay method can objectively reflect the differences in antioxidative activities of samples from fructus lycii.This might be used for quality control of this medicine and other antioxidative TCMs,as well as for high-throughput activity screening.
