维甲酸诱导肺腺癌细胞凋亡的实验研究

摘要 目的 探讨诱导分化剂全反式维甲酸(ATRA)对人肺腺癌细胞株A549 和SPCA1 的影响。 方法 实验分为不同浓度的维甲酸用药组及对照组,进行氚标记胸腺嘧啶脱氧核苷(3HTdR)掺入细胞生长测定,电子显微镜下观察细胞核形态及用流式细胞仪测定细胞DNA 含量,分别做凋亡细胞的定性和定量分析。 结果 ATRA 在10 μm ol/L浓度时即对肺癌细胞A549 和SPCA1的生长有明显的抑制作用,且呈剂量依赖性。此种抑制是通过诱导细胞凋亡达到的。A549 和SPCA1凋亡细胞率在50 μm ol/L药物浓度分别为3518% 和1920% 。 结论 ATRA 在体外可以诱导肺腺癌细胞的凋亡,为临床上通过诱导细胞分化促进和触发肿瘤细胞凋亡,从而为预防和治疗肿瘤提供了理论依据。 Objective At the end of their life span, differentiated cells die by apoptosis. There are many observations that subsets of differentiating cells undergo apoptosis relatively early after induction of cell differentiation. The purpose is to investigate the effect of all trans retinoid acid(ATRA),which triggers myelocytic differentiation, on human lung adenocarcinoma cell lines A549 and SPCA1. Methods A549 and SPCA1 cells were exposed to various concentrations of ATRA. 3H TdR incorporation, cell cycle analysis of flow cytometry and morphological observations with electron microscope were used to investigate cell growth condition and ascertain the mode of cell death. Results ATRA inhibited the growth of lung adenocarcinoma cells both A549 and SPCA1 cell lines in vitro. Drug inhibition was dose dependent. By using flow cytometry and morphological observation with electron microscope, it was found that the growth inhibition of the cells was caused by induction of cell apoptosis. At 50 μmol/L concentration of the drug, apoptotic cells of total cells were about 35 18% in A549 cells and 19 20% in SPCA1 cells. Conclusions The study suggests that growth retardation and regression of tumor achieved by enhancement of cell apoptosis after administration of differentiation inducing agents may be considered as a useful method in treatment and prevention of lung cancer.
