摘要
本研究以鸡肌苷酸合成代谢相关酶腺苷一磷酸脱氨酶(AMPD1)基因为候选基因,以宁夏地方品种固原鸡(白羽、麻羽、红羽),以及引进肉鸡品种AA鸡和蛋用品种罗曼鸡为研究对象,利用PCR-SCCP技术对AMPD1基因多态性进行了检测,并对其基因型、基因频率以及遗传特性进行了相关的统计.研究结果表明:AMPD1基因有2个等位基因(A、B),3种基因型(AA、AB、BB).固原鸡各品系BB基因型频率均高于AA鸡和罗曼鸡,尤其以固原鸡红羽为最高(0.822 2),与AA鸡之间存在着极显著的差异(P<0.01).对AMPD1基因的遗传特性统计分析结果表明,在所检测的几个品种鸡中杂合度均小于0.5.在多态信息含量中,固原鸡白羽、红羽在AMPD1基因多态位点上属于低度多态(PIC<0.25),其他各品种各多态位点多态信息含量(PIC)均处于中度多态(0.5>PIC>0.25).本研究结果将为宁夏地方品种固原鸡开发利用及肉质品质的改良提供重要的理论根据.
The adenylosuccinate lyase AMPD1 which participate in the process of inosine monophosphate anabolic metabolism,as candidate gene in the study,PCR-SSCP was used to detected Polymorphism AMPD1 gene on Guyuan Chicken of local variety in Ningxia and Arbor Acres chicken and Roman chicken of the introduced variety,and genotypic frequencies,allelic frequencies and genetic characteristics were calculated.The results of the analyses showed: The allele A and B,the genotype AA,AB and BB were detected on AMPD1 gene.The frequency of Guyuan Chicken with BB genotype was higher than Arbor Acres chicken and Roman chicken with BB genotype,and the genotypic frequency of Guyuan chicken(red plume) is the highest(0.8222),and the difference between Arbor Acres chicken was high significant Statistical results of the genetic characteristic of AMPD1 gene showed that the heterozygosis was less than 0.5 in the analytic chicken population.The value of AMPD1 gene in Guyuan Chicken(white plume),(red plume) possessed low Polymorphic information content(PIC0.25),however,the other breed had intermediate Polymorphic information content(0.5PIC0.25).The result would supply the theoretical reference for the exploitation and utilization of Guyuan Chicken in Ningxia and the improvement of meat quality of the chicken.
