摘要
目的 探讨内毒素性脑水肿降钙素基因相关肽(CGRP) 的表达及其意义。方法 30 只新西兰白兔,分内毒素组(25 只)和对照组(5 只) 。内毒素组于脑池内注射内毒素400 μg,对照组注入等容量生理盐水。于不同时间收集血液、脑脊液和脑组织,用放射免疫法测定CGRP含量,干燥法测定脑组织含水量。结果 注射内毒素后,脑组织( 海马区) 、脑脊液和血浆中CGRP含量显著高于对照组和注射前( P< 0-05 或P< 0-01) ,12~24 小时达高峰;脑组织含水量明显高于对照组( P< 0-05 或P<0-01),24 小时脑含水量达高峰。结论 内毒素诱导中枢神经系统炎症情况下,中枢和外周组织CGRP表达增强,CGRP过量的产生和释放可加剧脑水肿和炎症损害。
Objective To study the expression and significance of calcitonin gene related peptide (CGRP) in brain edema induced by endotoxin in rabbits Methods In 25 New Zealand white rabbits, 400 μg endotoxin was intraventricully injected. The same volume of normal saline was injected in 5 rabbits as controls. CGRP concentrations were detected by radioimmunoassay in different periods in the plasma, cerebrospinal fluid (CSF) and brain tissue (hippocampus area) The brain water content were measured by dry method Results The plasma, CSF and brain tissue CGRP concentrations after endotoxin injection were significantly higher than those of control group, and those before injection ( P< 0 05 or P< 0 01) The peak CGRP level occured on 12~ 24 hours after endotoxin injection Brain water levels were also significantly higher ( P< 0 05 or P< 0 01) and reached to the peak level on 24 hours after injection Conclusion The CGRP expression was increased in brain tissue, CSF and plasma during the inflammation of central nervous system induced by endotoxin Excessive CGRP production and release may worsen the process of brain edema and inflammation
