摘要
目的 阐明表皮生长因子受体 (EGFR)与人类骨髓造血细胞异常增生和异常发育的关系。方法 对5 6例恶性血液病、2 9例良性血液病及 9例正常人 ,应用 Southern Blot技术检测骨髓细胞中 EGFR基因的扩增与重排 ,应用免疫酶标技术检测骨髓细胞 EGFR的表达情况。结果 所检测的标本均未发现 EGFR基因的扩增及表达 ,仅在 2例急性白血病 (1例 M3 、1例 M6)中发现可疑重排带 ,2例重排带的大小相同均为 3 .3 kb。结论 EGFR基因的扩增与重排较少发生在人类血液系统恶性疾患中。人类良性和恶性骨髓造血细胞均无 EGFR的表达。提示EGFR表达异常可能较少与人类造血细胞的增殖与发育异常有关。在骨髓增殖异常综合征 (MDS)和白血病中可能存在发生扩增与异常表达的非 EGFR基因的其它与 v- erb B同源的基因 ,参与 MDS和白血病的发生与发展。
Objective To investigate the relationship between EGFR and bone marrow dyshemotopoiesis.Methods The bone marrow samples from 56 patients with malignant hemotologic disorders,29 patients with benign hemotologic disorders and 9 healthy controlls were detected for EGFR gene amplification and/or EGFR expression by using Southern blot assay with EGFRcDNA probe and by using immunochemical stains respectively.Results All of the cases determined have no EGFR gene amplification and EGFR expression.The rearrangements of the EGFR gene fragment were only found in 2 cases with acute leukemia (AL) including 1 case of M3 and 1 case of M6,in which the size of the rearranged fragments were the same 3.3kb.Conclusion 1.EGFR gene amplification and rearrangement rarely occur in human malignant hematologic disorders.2.It seems that the abnormal expression of EGFR may not be linked to dysproliferation and dysplasia of human hemotopoietic cells. 3.Our study combined with prior documents indicates that in MDS and AL may be exist other homologous genes of v erbB,which may be involved in the occurence and the development of hemologic malignant disorders.
