摘要
目的 探讨 Fasl(Fas- L igand)在 ABM(activated bone marrow)杀伤肿瘤细胞效应中的作用。方法 应用抗 CD3 单克隆抗体体外激活骨髓细胞 ,采用免疫荧光标记、流式细胞仪检测 ,观察了不同条件下 ABM细胞表面 Fas L 的表达。结果 未激活的骨髓细胞不表达 Fas L,r IL- 2激活后可见 Fas L 的表达 (7.0 4 %± 3 .4 4% ) ,抗 CD3单抗可促进其表达 (11.84 %± 4 .80 % ) ,三组比较具有显著性差异 (P<0 .0 0 5 )。结论 ABM杀伤肿瘤细胞的机制与 Fas/ Fas L 介导的肿瘤细胞凋亡有关。
Objective In order to study the effect of FasL expressed on bone marrow cells in killing tumor cells in vitro.Methods We observed the appearance of FasL under different occasions on the surface of activated bone marrow cells (ABM),from 20 patients with leukemia in complete remission state with the method of immunofluorescent sign and flowing cytometry. Results FasL can′t be observed until bone marrow are activated by rIL 2 (5.92%±4.86%),anti CD 3 monoclonal antibody can promoto the appearance (10.56±7.67%).Conclusion The remarkable difference of the three comparisons (P< 0.005) show the mechanism of ABM killing tumor cells related to the apoptosis mediated by Las/FasL.
