IFN-α和IL-2联合激活的脐血对K562细胞的杀伤和净化作用研究

STUDY ON KILLING AND PURGING EFFECT OF IL-2 CATIVATED CORD BLOOD IN COMBINATION WITH IFN-α ON K562 LEUKEMIC CELL

目的 探讨体外干扰素 α(IFN- α)联合白细胞介素 2 (IL- 2 )激活的脐血对慢性髓性白血病细胞株K5 62细胞的杀伤和净化作用。方法 采用细胞培养法和反转录聚合酶链反应法进行研究。结果 IL- 2激活 3 d的脐血对 K5 62细胞具有显著的杀伤和净化作用 ,加用 IFN- α后这种作用得到明显增强。IL- 2单用或与 IFN- α联合对脐血 CFU- GM未见明显抑制作用。结论 IFN- α可增强 IL- 2激活脐血对 K5 62的杀伤和净化作用。

Objective To evaluate the killing and purging effect of interleukin 2(IL 2)activated cord mononuclear cells(MNC) in combination with interferon alpha(IFN α) on K562 leukemic cells.Methods Semi solid colong culture and RT PCR technique were used.Results Cord blood MNC was activated in vitro with IL 2 for 3 days.The activated cord blood MNC displayed killing and purging effcet on K562 cells.This effect was further increased when IFN α added with IL 2.IL 2 alone or in combination with IFN α showed no significant inhibitory effect of CFU GM in cord blood.Conclusion IFN α can enhance the effect of the killing and purging of IL 2 activated cord blood MNC on K562 cells.
