摘要
目的 探讨基质金属蛋白酶2(MMP2) mRNA 及蛋白在胃癌中的表达。方法 用RTPCR 法及免疫组化法分别检测25 例胃癌患者癌组织及癌旁2cm 与≥5cm 组织MMP2 mRNA 及蛋白的表达。结果 胃癌组织MMP2 mRNA 表达率(17/25) 高于两组癌旁组织,与癌旁≥5cm 组织表达率(9/25)比较差异有显著性(P<0 .05);免疫组化显示胃癌组织MMP2 蛋白的表达率(17/25) 也高于两组癌旁组织,与癌旁≥5cm 组织表达率(8/25) 比较差异有显著性(P< 0 .05) 。结果接近RTPCR 法;胃癌组织MMP2 mRNA 表达值显著高于两组癌旁组织(P均<0 .01);MMP2 mRNA 及蛋白表达似与胃癌分化及临床分期有关。19 例进展期胃癌组织及癌旁2cm 组织,MMP2 mRNA 表达值显著高于6 例早期胃癌相应组织(P均< 0.05) 。结论 MMP2 在胃癌侵袭及转移过程中可能发挥重要作用。
Objective To investigate the expression of matrix metalloproteinase 2(MMP 2) mRNA and protein in gastric cancer. Methods Tumor tissues and adjacent normal tissues in 25 cases of gastric cancer were studied by using the reverse transcriptase polymerase chain reaction (RT PCR) and immunohistochemical techniques. Results The rate of expression of MMP 2 mRNA in tumor tissues was higher than that in 2cm and ≥5cm adjacent normal tissues and significantly higher than that in ≥5cm adjacent tissues ( P <0.05). The result of immnohistochemical staining was almost similar to that by RT PCR method. The signals of MMP 2 cDNA were significantly stronger in tumor tissues than those in the other two group of adjacent normal tissues ( P all<0.01). The mRNA expression correlated well with histological differentiaton and clinical staging of gastric cancer. The signals in tumor and in 2cm adjacent normal tissues of 19 cases with advanced cancer were significantly stronger than those in 6 cases with early cancer ( P all<0.05).Conclusions MMP 2 may play an important role in tumor invasion and metastasis in gastric cancer.
