摘要
对一株侧孢芽孢杆菌(BL2002)的产胞外酶(淀粉酶、蛋白酶、脂肪酶、纤维素酶)特性和其抑菌特性进行了初步研究,并对该菌株的产芽孢发酵条件进行优化。酶活鉴别培养基结果显示:BL2002具有较强的产胞外蛋白酶、纤维素酶、淀粉酶的能力,透明圈与菌落比各为3.74、5.89、5.60但未显示产脂肪酶的能力;采用平板交叉划线法研究其抑菌特性发现BL2002对7种病原菌(金黄色葡萄球菌、大肠杆菌、李斯特菌、沙门氏菌、小肠结肠耶尔氏菌、假单胞菌、肠球菌)有显著的抑制作用;优化后发酵条件为初始pH7.0、接种量4%葡萄糖2%、蛋白胨5%、CaCl20.05%、MnSO40.10%,该菌株此条件下在30 L罐中芽孢产量可达到2.39×109 cfu/mL。
This study investigated the extracellular enzyme and antimicrobial characters of a Bacillus laterosporus(BL2002).Its fermentation conditions was also optimized.Results showed that BL2002 is capable to select enzymes such as amylase,proteinase and cellulase but no lipase,ratio of clear zone and colony separately being 3.74、5.89、5.60.Besides,the antimicrobial activities of BL2002 were detected against 7 different pathogenic bacteria.The optimal fermentation conditions being initial pH7.0,inoculum 4 %,glucose 2 %,peptone 5 %,CaCl2 0.05 %,MnSO4 0.10 %,the spore yield with optimum medium formula in 30 L fermentor was up to 2.39×109 cfu/mL.
