摘要
利用PCR和DNA重组技术克隆了中国明对虾CrusFc-2基因的成熟肽编码序列并构建了pET-DsbA-CrusFc-2原核表达载体,转化E.coliBL21(DE3)后,经异丙基--βD硫代半乳糖苷(IPTG)诱导得到了重组蛋白.重组蛋白经纯化后免疫新西兰大白兔制备了抗血清,Western Blot分析表明CrusFc-2蛋白主要存在于对虾血细胞中.
The sequence coding mature peptide of CrusFc-2 was cloned from Chinese shrimp Fenneropenaeus chinensis,and a prokaryotic expression vector pET-DsbA-CrusFc-2 was constructed by PCR and recombinant DNA techniques.Fused protein was induced by IPTG in transformed E.coli BL21(DE3).The New Zealand white rabbits were immunized with the purified recombinant protein DsbA-CrusFc-2 to prepare antiserum.The results of western blot indicated that CrusFc-2 mainly existed in hemocyte of shrimp.
