摘要
为有效保护和利用华北大黄野生资源,以野生华北大黄为材料,探讨了不同种类、不同质量浓度的植物生长调节剂对其愈伤组织诱导及增殖效应,并采用高效液相色谱对愈伤组织及野生植株中土大黄苷含量进行了分析.结果表明:MS培养基中单独添加不同质量浓度的6-BA,不利于愈伤组织的增殖;培养基中单独添加不同质量浓度的NAA和2,4-D,对愈伤组织的增殖有一定作用;6-BA分别与低质量浓度NAA及2,4-D配合使用时,对愈伤组织的增殖效应十分明显.3种不同类型的愈伤组织中均检出土大黄苷,其中2种愈伤组织土大黄苷的质量分数高达1.45%~1.54%,为野生植株的3.63~3.85倍.
The effects of different plant growth regulators on callus proliferation were discussed,using wild plant of Rheum franzenbachii as materials in this study,and contents of rhaponticin in samples of callus and wild plants was analyzed by HPLC.The results showed that the MS medium plus different mass concentration of 6-BA lonely was not conducive to the proliferation of callus,and different mass concentration of NAA and 2,4-D lonely played a role for the proliferation.6-BA in combination with lower mass concentration of NAA or 2,4-D was very suitable for callus proliferation.Rhaponticin were detected in three types of callus,and the mass fraction of two types of calluses was as high as 1.45%~1.54%,3.63~3.85 times compared to that of wild plants.
