HPLC法测定当归流浸膏中阿魏酸和藁本内酯的含量

Determination of the Content of Ferulic Acid and Ligustilide in Chinese Extractum Angelicae Liquidum by HPLC

目的测当归流浸膏中阿魏酸和藁本内酯的含量,为当归的质量标准提供一定的依据。方法采用HPLC法测定不同厂家当归流浸膏中阿魏酸和藁本内酯的含量,实验以乙腈-0.085%磷酸液为流动相,采用梯度洗脱,流速:1.0 mL/min;柱温:25℃;检测波长:325 nm。结果不同厂家当归流浸膏中阿魏酸的含量在(0.2752～0.6094)mg/mL之间;藁本内酯的含量在(0.8186～5.8832)mg/mL之间;总固体在(3.4209～5.3632)g之间。结论不同厂家的当归流浸膏中阿魏酸、藁本内酯的含量差异较大,同一厂家不同批次的当归流浸膏中阿魏酸、藁本内酯的含量也有一定的差异,应尽快完善当归流浸膏的质量标准。

Objective To determine the content of ferulic acid and ligustilide of Chinese extractum angelicae liquidum from different manufacturers,and provide some academic basis for quality criterion of Chinese extractum angelicae liquidum.Methods Using the method of HPLC to determine the content of ferulic acid and ligustilide.This method was established with the mobile phase consisting of methanol acetonitril-0.085% phosphoric acid and flow rate of 1.0 mL/min and the temperature at 25 ℃,the detection wavelength was 325 nm.Results The content of ferulic acid was between 0.2752 ～ 0.6094 mg/mL;the content of ligustilide was between 0.8186 ～ 5.8832 mg/mL;the total solid content was between 3.4209 ～ 5.3632 g.Conclusion The contents of two kinds of active compositions are significantly different in different manufacturers of Chinese extractum angelicae liquidum,so we should improve the academic basis of quality criterion for Chinese extractum angelicae liquidum.
