摘要 目的建立一种灵敏、快速的实时荧光PCR检测系统对HBV进行筛查和准确定量。方法采用本研究建立的实时荧光PCR检测对30例慢性HBV患者的血样和70份献血员血样进行HBV检测并将30例HBV阳性样本进行基因分型测定,与一种商品化的检测进行对比。结果采用WHO国际标准品来评价该检测系统的检测灵敏度,批内和批间变异系数(CV)分别为1.4%-9.4%,0.0%~2.3%。与上海科华HBV核酸检测系统的检测结果相关性良好(r=0.95,P〈0.0001)。结论:该检测系统可以对血浆中的HBV进行检测以及准确定量,可以用来筛查献血员以及进行HBV、HIV和HCV多重检测,并进行大规模的应用。 Objective To establish a sensitive and rapid real- time fluorescence PCR detection system for screening and quantification of HBV. Methods 30 blood samples of patients with chronic HBV and 70 blood donors samples were detected the established real - time fluorescence PCR detection system. 30 cases of HBV - positive samples were HBV genotype determined with a commercial test to compare. Results The WHO international standard to evaluate the sensitivity of the detection system, inter - and intra coefficient of variation (CV) 1.4% - 9. 4%, 0.0% - 2.3%. China and the Shanghai Branch of the HBV DNA detection system test results correlated well (r= 0.95, p 〈 0.0001). Conclusion The detection system can be detected in plasma, and quantification of HBV can be used to screen bloed donors as well as HBV, HIV and HCV multiple testing, and large - scale applications.
