摘要
本研究旨在通过移植肿瘤动物模型来现察软坚消肿液的抑瘤作用,并探讨该药的急性毒性反应。采用孙氏综合法测定药物LD_(50),移植S(180)肿瘤细胞及人胃癌细胞(MGC—803)建立小鼠肿瘤模型,测定肿瘤重量了解药物抑瘤情况,透射电镜现察肿瘤细胞变化,并通过Northern杂交观察癌基因Bcl—2、C—myc和P_(53)的改变。该药腹腔注射的LD_(50)为29.34±2.49g/kg;灌胃的LD_(50)为125.34±20.53g/kg。软坚消肿液对S_(180)肿瘤的最大抑瘤率为38.4%,对人胃癌移植肿瘤的最大抑瘤率为56.01%,透射电镜可见复方中药处理的小鼠,瘤细胞呈现典型凋亡细胞特征,Northern杂交显示癌基因Bcl—2表达下调,C—myc和P_(53)表达增强。研究表明软坚消肿液对动物移植肿瘤有明显的抑制作用,可诱导S_(180)肿瘤细胞和人胃癌细胞凋亡。该药的毒副作用极小,有临床开发应用价值。
Objective of the study is to observe antineoplastic effect of the herbal medicinal mixture on the animal model of implanted tumor and to explore the acute toxicity of the mixture. Sun's comprehensive method was used to explore LD50 of the mixture to set up the animal model of S180 Sarcoma and human gastric cancer to survey tumor weights for undertanding antineoplastic effect of the mixture,adopted transmission electron microscopy (TEM),and Northern blot to detect the genetic expression. LD50 of the mixture is repectively 29. 34±2. 49g/kg(ip) and 125. 34±20. 53g/kg(drench). The antineoplastic rate of mixture is 38. 4% (for the S180 sarco-ma)and 56. 1%(for the human gastric cancer)respectively. The tumor cells from the mice treated with the mixture were displayed typical changes of apoptosis by TEM. with Northern blot, the expression of Bel-2 was reduced and that of C-myc and P53 wre enhanced compared to the controlled group respectively. The herbal medicinal mixture is able to restrain inplanted tumor to grow in mice,and to induce apoptosis of S180 sarcoma cells and human gastric carcinoma cells. The toxicity of the mixture is very little. The herbal medicinal mixture has good clinical use Value
