摘要
目的 采用脾脏分段灌洗与保存的方法来选择一种适合于脾脏的灌洗液。方法 根据脾脏的解剖学分段将脾灌洗后, 通过不同时限灌洗脾段的细胞存活率及光、电镜观察细胞形态, 比较Hartmanns 液、Collins 液和WMOI号液的优劣。结果 根据三种检测指标, WMOI号液保存脾的细胞存活率明显高于其它二种( P< 0.01), 光、电镜检查细胞存活时限亦为WMOI号液明显高于其它二种( P< 0.01 或P< 0 .05) 。结论 认为在这三种灌洗液中以WMOI号液最适于脾脏。
Objective To choose the solution suitable for spleen transplantation by using method of splenic segmental perfusion and preservation.Methods After the splenic artery and vein were dissociated clearly, three kinds of perfusion solutions (Hartmanns solution, Collins solution and WMO I solution) were used to randomly perfuse the corresponding segments of spleen. When the efferent fluids from the splenic vein became clear, the perfused spleen segments were preserved for different duration with the same perfusing solution to measure the survival rate of splenocytes (SRS) and the cell morphology was observed under light and electron microscopy.Results Among the three solutions, SRS with WMO I solution was significantly higher than that of the other two solutions ( P < 0.01 ). The perfused spleen with WMO I solution showed the milder morphological changes and a significantly longer preservation duration than those with other two solutions ( P < 0.05 or P < 0.01 ).Conclusion Among the three solutions, WMO I solution was most suitable for perfusion and preservation of spleen.
