摘要
目的观察芪棱汤对脑缺血再灌注损伤后兴奋性氨基酸(EAA)的影响,为益气养阴活血法治疗缺血性中风提供实验依据。方法将大鼠随机分为假手术组、模型组、补阳还五汤组及芪棱汤组。采用颈内动脉线栓加环扎法复制大鼠脑缺血再灌注模型。于再灌注后不同时间段观察大鼠缺血脑组织的谷氨酸(Glu)、门冬氨酸(Asp)的含量。结果模型组再灌注后各时间段的Glu含量及再灌注后24h段的Asp含量较假手术组显著减少,补阳还五汤组在再灌注24h段的Glu含量较模型组升高,芪棱汤组在再灌注24h段的Glu、Asp含量较模型组均明显升高,且芪棱汤组在再灌注24h段升高Glu、Asp方面优于补阳还五汤组。结论芪棱汤可降低EAA的神经毒性,从而对脑缺血再灌注所导致的脑损伤起保护作用。
Objective:To explore the influence of Qileng Decoction on expression of excitatory amino acid after focal cerebral ischemic reperfusion in rats and to provide the scientific foundation for benefiting vital energy,nourishing yin and activating blood circulation on ischemic stroke.Methods:The rats were randomly divided into 4 groups including the sham-operation,the model group,the Buyanghuanwu Decoction group and the Qileng Decoction group.The model of focal cerebral ischemic reperfusion injury was made by using thread embolizing and fastening of the ICA in rats.Content including Glu,Asp and GABA were measured by high-performance liquid chromatography at different time point after cerebral ischemic reperfusion.Results:The content of Glu at every time point and the content of Asp at 24 hours after reperfusion in model group were all significantly lower than those in the sham-operation group.The content of Glu in Buyanghuanwu Decoction group was increased compared with that in the model group at 24 hours after reperfusion.Content including Glu and Asp in the Qileng Decoction group were increased significantly compared with those in the model group at 24 hours after reperfusion.Contrasted with Buyanghuanwu Decoction group,the contents of Glu,Asp in Qileng Decoction group were increased remarkably at 24 hours after reperfusion.Conclusion:Qileng Decoction plays the nerve protective effect by reducing the nerve toxicity of EAA.
