摘要
目的研究Bcl-2、Bax在CCl4诱导小鼠急性肝损伤过程中的动态变化,并探讨其作用机制。方法试验共分10组:CCl4腹腔注射(浓度0.1%溶于橄榄油中,0.1 mL/10 g)诱导小鼠急性肝损伤3、6、12、24、30、36、42、48、54 h及正常小鼠。观察各组小鼠肝脏组织病理变化;用Western blotting法检测各组肝脏Bcl-2、Bax蛋白的表达以及变化趋势。结果肝损伤之后,小鼠肝脏失去其正常结构,36 h肝脏病理改变最为显著。CCl4诱导小鼠急性肝损伤3、6、12、24、30、36、42、48、54 h之后肝脏Bcl-2和Bax蛋白的表达均呈明显变化趋势(P<0.05);Bcl-2在CCl4处理后明显降低,在24 h至最低点,然后开始升高但未恢复正常,至48 h和54 h又再次降低;Bax在CCl4处理后先升高,在12 h达到高峰,然后降低,并逐渐向正常恢复。结论 CCl4诱导小鼠急性肝损伤过程中,Bcl-2和Bax蛋白表达显著变化,凋亡和抗凋亡因素在CCl4诱导小鼠急性肝损伤过程中可能起到重要作用。
Objective To study the dynamic changes of the expression of Bcl-2 and Bax during acute liver injury of mice induced by CCl4. Methods The acute liver injury of mice was induced by CCl4(0.1% in olive oil,0.1 mL/10 g) by abdominal injection.The liver histopathological changes were observed at 3,6,12,24,30,36,42,48,54 h after CCl4 injection.The protein expression of Bcl-2 and Bax were detected by Western blotting during the process. Results After CCl4 administration,the livers' structures were all abnormal.At 36 h,the liver injury reached the highest level.The protein expression level of Bcl-2 and Bax were significantly changed in the liver of mice during acute liver injury(P0.05).The expression of Bcl-2 was significantly decreased after CCl4 injection.It decreased to the lowest levels at 24 h.After 24 h,the expression of Bcl-2 was increased but did not reached to the normal level.From 48 h to 54 h,it decreased again.The protein expression of Bax was significantly increased after CCl4 injection and it increased to the highest levels at 12h.Then it decreased to the normal level gradually. Conclusion During acute liver injury induced by CCl4,the protein expression of Bcl-2 and Bax is significantly changed.Apoptosis factors and anti-apoptosis factors probably play important roles in acute liver injury induced by CCl4.
