蛤蚧肽对荷瘤小鼠的免疫调节及抗肿瘤作用 被引量:13

摘要 目的观察蛤蚧肽对S180和Hepa1-6荷瘤小鼠免疫调节及其对小鼠移植肿瘤的生长抑制作用。方法以S180和Hepa1-6荷瘤小鼠为模型,研究蛤蚧肽及其联合环磷酰胺(CTX)对肿瘤的治疗效果,采用MTS法测定小鼠腹腔巨噬细胞杀瘤活性、脾淋巴细胞刺激指数、自然杀伤细胞(NK)活性,并称瘤重,计算抑瘤率。结果单纯蛤蚧肽灌胃400 mg.kg-1.d-1,连续12 d,可显著提升S180荷瘤小鼠的腹腔巨噬细胞杀瘤活性及Hepa1-6荷瘤小鼠的腹腔巨噬细胞吞噬功能(P<0.05),蛤蚧肽治疗后S180及Hepa1-6荷瘤小鼠脾淋巴细胞增殖能力及NK细胞活性均显著提高(P<0.05)。与CTX联合应用时,蛤蚧肽能使受CTX抑制的上述免疫指标得到改善,并能显著提高抑瘤率。结论蛤蚧肽对肿瘤及化疗药物造成的免疫功能抑制有调节作用,并可通过此途径达到协同化疗药物抗肿瘤效果。 Objective To study the immunoregulation and growth inhibitory effects of gecko peptides in tumor-bearing mice.Methods The mice with sarcoma-180(S180) and Hepa1-6 hepatic carcinoma as the animal models were used to study the therapeutic effect of gecko peptides united with CTX.And MTS was applied to determine tumoricidal activity of peritoneal macrophage,the stimulation index of lymphocyte and natural killer cell activity.Then the tumor weight and the inhibiting tumor rate were calculated.Results It can significantly enhance the tumoricidal activity and phagocytosis(P0.05) of peritoneal macrophage of the mice with S180 and Hepa1-6 hepatic carcinoma,giving gecko peptides 400 mg.kg﹣1 alone,for 12 days,it can also enhance the proliferation capacity of lymphocyte and activity of natural killer cells remarkably(P0.05) after treatment with gecko peptides.United with CTX,gecko peptides can improve above immune indexes which were constrained by CTX,and can enhance the tumor inhibition rate significantly(P0.05).Conclusion Gecko peptides can improve the suppression of immune function caused by tumour and chemotherapeutics,and can achieve its anti-tumor effects cooperated with chemotherapeutics.
