摘要
目的:建立高效液相色谱法测定对硝基苯-α-D-吡喃葡糖苷的含量。方法:采用Hypersil C18色谱柱(250 mm×4.6mm,5μm);流动相-甲醇-水(759∶25),流速为0.8 mL·min-1,紫外检测器:检测波长303 nm,柱温为35℃。结果:对硝基苯-α-D-吡喃葡糖苷的浓度在13.5~135.0μg·mL-1的范围内,峰面积与浓度呈良好线性(r=0.9999)。方法平均回收率为99.7%(n=6)。结论:本法简便、快速、准确,可用于对硝基苯-α-D-吡喃葡糖苷的含量测定。
Objective:To establish an HPLC method for determination of P-nitrophenly-α-D-glucopyranoside(α-PNPG).Methods:α-PNPG was determinated by HPLC with a Hypersil C18(column,250 mm×4.6 mm,5 μm)with UV detection at 303 nm,the mobile phase was contained of methanol-water(75∶25),the flow rate was 0.8 mL·min-1,the column temperature was 35 ℃.Results:The calibration curve was linear in the range of 13.5-135.0 μg·mL-1(r=0.9999) for α-PNPG.The average recovery was 99.7%(n=6).Conclusion:The method is simple,rapid,accurate,reliable,high sensitivity and good separation,which may be used for the determination of α-PNPG.
