摘要
采用柴胡、当归代替卡介苗(BCG)、以新西兰纯种白兔成功地制备了具有抗癌效应的肿瘤坏死因子(TNF)。获得的TNF阳性血清(TNS)经60%饱和硫酸铵沉淀、Sephadex G-200凝胶过滤和制备性聚丙烯酰胺凝胶电泳(PAGE)等三步纯化,活性提高27倍。以肝癌细胞作为靶细胞,加入适量TNF对癌细胞进行毒性效应试验,结果癌细胞坏死、裂解;用Hela细胞和肺腺癌细胞作同样试验,亦获相同结果。
Tumor necrosis factor, TNF ( cachectin ) is a metabolically active protein secreted by macrophages in response to certain invasive stimuli, e.g. BCG and LPS. The authors prepared highly potent TNF by using Bupleurum chinense or Angelica sinensis as the initial stimulant instead of BCG. The TNF-positive serum thus obtained was purified in 3 steps: first by 60% saturation of (NH_4)_2SO_4 fractionation, followed by Sephadex G-200 gel filtration and finally preparative polyacrylamide gel electrophoresis, resulting 27-fold elevation of the activity. Cytotoxicity test performed by addition of suitable amount of the purified TNF to liver cancer cellscauses necrosis and cytolysis of the cancer cells. Hela cells and lung adenocarcinoma cells also are prone to the cytolytic effect of the TNF preparation.
