生精散治疗大鼠弱精子症的实验研究 被引量:12

摘要 目的研究生精散对大鼠弱精子症的治疗作用。方法采用雷公藤多甙制备弱精子症大鼠模型,通过计算机辅助精子分析系统观察生精散对精子活率、活力、密度以及各种运动速度和运动方式等功能指标的影响。实验动物按体重随机分为正常对照组、雷公藤多甙组、生理盐水组及生精散小、中、大剂量组,每组5只。正常对照组每日给予正常饮食,共42 d;生理盐水组给予生理盐水1 mL/d,共28 d;雷公藤多甙组每日灌胃雷公藤多甙20 mg/kg,共4周;生精散小[0.01 g/(只.d)]、中[0.02 g/(只.d)]、大[0.03 g/(只.d)]剂量组灌胃生精散,共14 d。结果雷公藤多甙组大鼠精子的活率、活力、密度以及直线运动速度(VSL)、平均路径速度(VAP)和曲线运动速度(VCL)等功能指标均较正常对照组和生理盐水组显著降低。睾丸组织HE染色显示,雷公藤多甙组大鼠生精小管内各级精母细胞和精子细胞数明显减少,生精细胞排列紊乱。生精散大、中、小剂量组均可显著提高模型大鼠精子的活率、活力、密度和运动速度(VSL、VAP、VCL);而对精子的运动方式,生精散大剂量组可以改善鞭打频率和平均移动角度。结论雷公藤多甙可以诱导稳定的弱精子症大鼠模型,生精散具有提高弱精子症大鼠精子的密度、活力、活率及运动速度的作用。 Objective To study the effect of Shengjingsan recipe on asthenozoospermia in rat model. Methods Rat model of asthenozoospermia was induced by oral administration of multi-glycosides of tripterygium wilfordii (GTW) everyday for 28 days. The sperm viability and the sperm vitality, sperm concentration as well as the sperm mobility were examined by computer assisted sperm analysis (CASA), and the sperm morphology was detected by hematoxylin-eosin (HE) staining. Rats were randomly divided into normal control group (normal diet, 42 d), NS group (1 mL/d NS, 28 d), GTW group (20 mg/kg GTW, 4 weeks) and small (0.01 g/d)-, medium (0.02 g/d)-, large (0.03 g/d)-dose of Shengjingsan Recipe group (14 d), five rats in each group. Results The parameters of the sperm viability and the sperm vitality, sperm concentration as well as the sperm mobility including straight-fine velocity (VSL), curve-line velocity (VCL), and average path velocity (VAP) decreased significantly in GTW group compared with normal control group and NS group. The HE staining of the testicle tissues showed that the spermatocytes and the spermatozoon cells reduced obviously in the seminiferous tubule in GTW group, in which abnormal arrangement of the spermatogerdc cells occurred. The above decreased parameters of the sperm viability and the sperm vitality, sperm concentration as well as the sperm mobility, induced by GTW, could be reversed effectively after Shengjingsan recipe treatment at small, medium, and large dosage. Large dose of Shengjingsan recipe ameliorated beat-cross frequency and mean angular displacement of sperm. Conclusions GTW can induce typical asthenozoospermia in rats. Sheng^ingsan recipe has therapeutic effect on asthenozoospermia by improving the sperm viability and the sperm vitality, sperm concentration as well as the sperm mobility.
