自发性高血压大鼠主动脉中一氧化氮/一氧化氮合酶体系的变化 被引量:6

Alteration in Nitric Oxide/Nitric Oxide Synthase System in Aortas of Spontaneously Hypertensive Rats

摘要 目的探讨自发性高血压大鼠主动脉中一氧化氮(NO)/一氧化氮合酶(NOS)体系的变化。方法随机选取健康雄性4周龄Wistar大鼠7只和4周龄自发性高血压大鼠7只,分别作为正常对照组及高血压组。4周龄及12周龄时检测二组大鼠血压。8周龄时取2组大鼠主动脉组织,采用硝酸还原酶法测定其NO水平,Western blot法检查其内皮型NOS(eNOS)以及诱导型NOS(iNOS)蛋白水平。用SPSS13.0软件对数据进行统计学分析。结果 12周龄时高血压组大鼠血压明显高于正常对照组[(24.1±0.5)kPavs(15.9±0.3)kPa](P<0.05)。12周龄时高血压组大鼠主动脉组织中NO水平较正常对照组主动脉中NO水平低[(7.2±1.7)μmol.(g prot)-1vs(17.1±5.3)μmol.(g prot)-1](P<0.05)。12周龄时高血压组大鼠主动脉中eNOS蛋白表达量与正常对照组大鼠比较差异无统计学意义[(0.5±0.2)vs(0.5±0.3)](P>0.05);12周龄时高血压组大鼠主动脉中iNOS蛋白表达量显著低于正常对照组大鼠[(0.9±0.3)vs(1.5±0.5)](P<0.05)。结论自发性高血压大鼠主动脉中NO/NOS体系下调参与高血压的形成过程。 Objective To investigate the alteration in nitric oxide(NO)/nitric oxide synthase(NOS) pathway in aortas of sponta-neously hypertensive rats(SHR).Methods Seven male Wistar rats of 4 weeks were randomly assigned to control group.Seven male SHR were served as SHR group.At 4 weeks and 12 weeks old,the blood pressure of rats in control group and SHR group were determined.At 12 weeks,the aorta of rats in the 2 groups were collected for testing NO production,eNOS and iNOS protein expressions were determined by Wes-tern blot method.SPSS 13.0 software was used to analyze the data.Results When rats become 12 weeks old,the blood pressure in SHR group increased significantly compared with that of control group [(24.1±0.5) kPa vs(15.9±0.3) kPa](P0.05).NO production in the aortic tissue of SHR group was significantly lower than that in control group [(7.2±1.7) μmol·(g prot)-1 vs(17.1±5.3) μmol·(g prot)-1](P0.05).eNOS protein expression in the aortic tissue of SHR group had no significant difference compared with that in control group [(0.5±0.2) vs(0.5±0.3)](P0.05).iNOS protein expression in the aortic tissue of SHR group was significantly lower than that in control group [(0.9±0.3) vs(1.5±0.5)](P0.05).Conclusion The down-regulation of NO/NOS system in SHR aorta is involved in the development of hypertension.
