摘要
目的探讨细丝蛋白A(filamin A,FLNa)在食管鳞癌组织中的表达及其临床意义。方法采用免疫组织化学方法(SP法)和逆转录聚合酶链反应(reverse transcription polymerase chine reaction,RT-PCR),分别检测32例食管鳞癌组织及距食管癌组织边缘5 cm以上的镜下未见癌浸润的正常组织中FLNa蛋白、FLNa mRNA的表达情况。结果在食管鳞癌组织及正常食管组织中,FLNa蛋白的表达阳性率分别为34.4%、84.4%,FLNa mRNA的表达量分别为0.376±0.053、0.517±0.082,FLNa蛋白明显降低(P<0.05)。结论食管鳞癌组织中FLNa蛋白和FLNa mRNA的表达均降低,可能与食管癌的发生、发展相关。检测FLNa的表达能对食管癌的临床治疗和预后判断有一定价值。
Objective To investigate the expression of filamin A(FLNa) in esophageal carcinoma tissues and its significance.Methods Immunohistochemistry(SP) and RT-PCR were respectively used to detect the expression of FLNa in 32 specimens of esophagel squamous cell carcinoma and normal esophagel tissues(5 cm distance away from the margin of esophagel cancer and no cancer infitration in microscope).Results The positive rates of FLNa protein in esophagel carcinoma tissues and normal tissues were 34.4% vs 84.4%(P0.05).The relative contents of FLNa mRNA were 0.376±0.053,0.517±0.082 in esophagel carcinoma tissues and normal tissues,the expressions of esophagel carcinoma tissues showed significant decreases(P0.05).Conclusion The expression of FLNa protein and FLNa mRNA are both lower in esophagel carcinoma tissues,suggesting that FLNa gene may be closely associated with oncogenesis,malignant degree.Detection of FLNa may be used for early diagnosis,clinical therapy and prognostic judgement of esophagel carcinoma.
