摘要
目的探讨电针对脑缺血再灌注模型大鼠谷胱甘肽抗氧化能力的影响。方法用改良Longa氏线栓法制备脑缺血再灌注大鼠模型,电针大椎、百会穴,比色法测定血清谷胱甘肽(GSH)含量及谷胱甘肽过氧化物酶(GSH-Px)、谷胱甘肽还原酶(GR)活性。结果模型组大鼠的GR、GSH-Px活性与GSH含量较假手术组大鼠均明显下降,电针大椎、百会穴后GR、GSH-Px活性与GSH含量均明显增加,但GR、GSH-Px活性仍明显低于假手术组,GSH含量接近假手术组,而在电针+MAPK通路阻断剂PD98059作用,大鼠GR、GSH-Px活性、GSH含量与模型组相比无显著性差异。结论电针可减轻脑缺血再灌注损伤而发挥整体抗氧化保护作用,其机制与调控机体内GSH-Px及GR的活性,促进机体谷胱甘肽系统保持一较高的动态平衡有关,亦可能与MAPK信号通路有关。
OBJECTIVE To investigate the effects of electro-acupuncture(EA) on glutathione oxidation in cerebral ischemia-reperfusion model rats.METHODS Longa suture-occluded method was adopted to create cerebral is chemia-reperfusion model rats which were then electro-acupunctured on GV-14 and GV-20 points and tested by chromatometry to know the contents of GSH as well as the activity of GSH-Px and GR.RESULTS Both the contents of GSH and the activity of GSH-Px and GR in EA treated group were significantly declined compared with those in sham-operated group.Electro-acupuncture on GV-and GV-20 points activated GR and GSH-Px which though were still lower than those in sham-operated group and increased the amount of GSH which was close to that of sham-operated group.Besides,the above indexes showed no great difference between EA treated and sham-operated groups when electro-acupuncture was companied with MARK pathway inhibitor PD9805.CONCLUSION Electro-acupuncture can reduce the damage caused by cerebral ischemia-reperfusion so as to display its comprehend antioxidation protection,whose mechanism may due to adjusting the activity of GSH-Px and GR,preserving the dynamic balance of glutathione system or MAPK signal channel.
