摘要
目的:探讨烧伤患者血清对体外培养的骨髓间充质干细胞的生物学行为的影响。方法:培养BM-MSCs,传代后分别用含有体积分数为10%正常人血清、烧伤患者血清的低糖达氏修正依氏培养基培养,用噻唑蓝(MTT)法测定其生长曲线,碘化丙啶(PI)染色,流式细胞仪分析细胞周期、β-gal染色测定衰老的MSCs。结果:含正常血清培养基传代培养的细胞生长曲线较烧伤患者血清慢,烧伤患者血清处理的细胞中G0/G1期细胞比例明显低于正常血清组(P<0.01),β-gal染色阳性细胞数目显著低于正常血清组。结论:烧伤患者血清比正常血清能更好地促进BM-MSCs生长;烧伤血清内可能含有多种能影响BM-MSCs生长的物质。
Objective:To investigate the effect of serum from normal and burned patients on the biological behaviors of bone marrow mesenchymal stem cells(BM-MSCs).Methods:BM-MSCs were incubated in L-DMEM in the presence of normal serum and serum harvested from burned patients after burn injury.The growth curve,cell cycle,β-gal positive cells of BM-MSCs were investigated.Results:The growth curves showed that the growth of MSCs in normal was slower than that in burned patients serum(P0.01).The ratios of cells at phase G0/G1 in the group of burned serum were significantly lower than those in the normal groups(P0101),but the ratios of β-gal positive cells were adverse(P0.01).Conclusion:Burned serum is more effective to stimulate proliferation of BM-MSCs than normal.There are probably many factors affecting the growth of BM-MSCs.
