姜黄素、小檗碱及其配伍对db/db小鼠糖脂代谢相关基因mRNA表达的影响 被引量:5

摘要 [目的]观察姜黄素、小檗碱分别及其配伍对db/db小鼠糖脂代谢相关基因mRNA表达的影响,为中药配伍合理性提供实验依据。[方法]db/db小鼠分别及联合口服给予姜黄素和小檗碱,给药28 d,实时定量逆转录聚合酶链反应(RT-PCR)法检测肝脏中葡萄糖-6-磷酸酶(G-6-Pase)、低密度脂蛋白受体(LDLR)和血管生成素相关生长因子(AGF)的mRNA表达。[结果]将小檗碱和姜黄素分别及配伍作用于db/db小鼠,姜黄素低剂量(19.5mg/kg)小檗碱高剂量(144mg/kg)配伍组可下调db/db小鼠肝脏G-6-PasemRNA表达,姜黄素高剂量(39 mg/kg)小檗碱高剂量(144 mg/kg)配伍组可上调db/db小鼠肝脏LDLR mRNA表达,各组对db/db小鼠肝脏AGF mRNA表达均无显著性影响。[结论]小檗碱和姜黄素配伍后,可调节糖脂代谢相关基因mRNA的表达,其作用优于单独给药组。 [Objective] To observe the effects of curcumin,berberine and its compatibility on mRNA expression of glucose and lipid metabolismrelated genes in db/db mice and provide the experimental basis for the rational com patibility.[Methods] The db/db mice were orally administered with curcumin and berberine for 28 days.Real Time RTPCR was used to detect G6Pase LDLR and AGF mRNA expression in liver.[Results] Treating db/db mice with curcumin,berberine and its compatibility,curcumin lowdose(19.5 mg/kg),berberine highdose(144 mg/kg) could downregulate the expression of LDLR in liver,but without significant effect on AGF mRNA.[Conclusion] Compatibility of berberine and curcumin,may regulate the expression of glucose and lipid metabolismrelated genes mRNA and its effects are better than the separated administration group.
