大肠杆菌O157∶H7胶体金试纸的制备与检测

Preparation of gold-immunochromatography strip for rapid detection of E.coli O157:H7

目的:制作大肠杆菌O157:H7胶体金金标试纸,检测试纸的灵敏度和特异性效果。方法:利用胶体金免疫层析技术,采用双抗体夹心法检测大肠杆菌O157,并对该法进行敏感性、特异性效果进行评估。结果:该试纸可在15min内检测大肠杆菌O157,灵敏度最低检测浓度为1×106cfu/mL。对于不同的肠道菌群(沙门氏菌、单增李斯特菌、空肠弯曲杆菌、副溶血弧菌),未发现有交叉反应,特异性良好。结论:大肠杆菌O157:H7金标试纸检测速度快、灵敏度高,适用于快速检测。

Objective：Make gold-immunochromatography strip for Escherichia coli O157：H7 and test the effect of sensitizer.Methods：Use the gold immunochromatography assay（GICA） technology,the Escherichia coli O157：H7 was detected by double antibody sandwich method.Results：This gold-immunochromatography kits can checkout Escherichia coli O157：H7 in 15 minutes.The sensitivity of the test is 1×106 cfu/mL.For different parasitism population,such as Salmonella,Vibrio parahaemolyticus,Campylobacter jejuni,not find cross-reaction.Indicating good specificity of this strip.Conclusion：The gold-immunochromatography strip is test quick,sensitivity high and well,apply to the express test.
