摘要
制备了玉米赤霉烯酮人工抗原,并且制备抗血清,为进一步建立酶联免疫分析方法奠定基础。应用活泼酯法和混合酸酐法分别合成人工免疫原和人工检测原,进行动物免疫获得抗血清。结果表明,经紫外吸收、红外光谱、荧光光谱和酶联免疫方法验证人工抗原合成成功,玉米赤霉烯酮-牛血清白蛋白、玉米赤霉烯酮-卵清蛋白偶联比分别为12:1和8:1,抗血清效价可达到102 400。合成的玉米赤霉烯酮免疫原具有良好免疫活性,检测原具有灵敏的检测性,为进一步建立玉米赤霉烯酮免疫学分析方法提供依据。
In order to build ELISA method for zearalenone,immunogenic antigen(ZEN-BSA)and coationg antigen(ZEN-OVA)of zearalenone(ZEN) were prepared,and then antiserum was produced.Through the carbodiimides reaction,ZEN was linked to bovine serum albumin(BSA)to generate immunogenic antigen ZEN-BSA and it also was linked to ovalbuminto(OVA) to generate coating antigen ZEN-OVA.To get antiserum,New Zealand white rabbits were immunized with ZEN-BSA.The products were identified through UV absorption,fluorescence spectroscopy,infrared spectra,and ELISA.The coupling ratio was 12:1 and 8:1 for ZEN-BSA and ZEN-OVA,respectively.And the titration of antiserum was 102 400.The results suggest that the immunogenix antigen can induce the generation of specific antibody of ZEN,and they also show good sensitivity of ZEN-OVA.The properties of ZEN artificial antigen make it applicable to immunoassay.
