苯中毒再生障碍性贫血患者血清对造血干/祖细胞集落形成及Fas表达影响 被引量:5

Effect of benzene poisoning aplastic anemia serum on hematopoietic stem/progenitor cell clone forming and Fas expression

摘要 目的观察苯中毒再生障碍性贫血(BPAA)患者血清中造血负调控因子对正常骨髓造血细胞的影响。方法以BPAA患者为研究对象,用其血清培养健康者骨髓细胞,进行骨髓造血干/祖细胞集落形成单位(CFU)计数,流式细胞仪检测CD34+细胞率及其Fas表达率的变化,同时用酶联免疫吸附法(ELISA)检测血清中干扰素-γ(IFN-γ)、肿瘤坏死因子-α(TNF-α)、白细胞介素-2(IL-2)的水平。结果 BPAA患者血清培养后骨髓粒-巨噬细胞集落形成单位(CFU-GM)和红系集落形成单位(CFU-E)数量均显著降低(均P<0.05),骨髓CD34+细胞比例显著降低,CD34+细胞的Fas表达率随之显著升高(均P<0.05);血清中IFN-γ、TNF-α、IL-2的水平也明显升高(均P<0.05)。结论 BPAA患者血清中造血负调控因子水平升高,使正常骨髓造血干/祖细胞集落形成受抑制,并上调造血干/祖细胞Fas表达。 Objective To study the effect of hematopoietic negative regulatory factors in benzene poisoning aplastic anemia(BPAA) serum on normal bone marrow hematopoietic cells.Methods Bone marrow cells from the healthy were incubated with BPAA serum,then colony-forming unit of bone marrow hematpoietic stem/progenitor cell was counted by microscopy,percentage of CD34+ cells and CD34+ Fas+ cells were analyzed by flow cytometry,serum interferon gamma(IFN-γ),tumor necrosis factor α(TNF-α),and interleukin 2(IL-2) levels were detected by enzyme-linked immunosorbent assay(ELISA).Results After incubated with BPAA serum,the numbers of bone marrow colony-forming unit-granulocyte and macrophage(CFU-GM),colony-forming unit-erythrocyte(CFU-E),as well as the percentage of CD34+ cells decreased significantly(P0.05),while the expression of Fas in CD34+ cells increased(P0.05).The concentrations of IFN-γ,TNF-α and IL-2 in BPAA serum were significantly higher than those of the healthy people(P0.05).Conclusion The levels of hematopoietic negative regulatory factors in BPAA serum raise,which inhibits hematpoietic stem/progenitor cells colony-forming and up-regulates the expression of Fas.
