摘要
制备含有五味子乙素的脂质体,用于运载抗癌药物阿霉素,即五味子乙素-阿霉素脂质体。脂质体对阿霉素的包封率用柱层析法测定,脂质体的形态、粒度分别用扫描电镜、激光粒度测定仪来观察与测定。采用昆明种小白鼠以及S-180动物肿瘤模型,评价五味子乙素-阿霉素脂质体的毒性与抗肿瘤作用。结果表明:五味子乙素-阿霉素脂质体的包封率可达(96.45±1.86)%,粒度范围为(153.2±19.6)nm,Zeta电位为(-38.6±2.5)mV,显著降低了阿霉素的急性毒性,同时显著提高了阿霉素的抗肿瘤作用。
To prepare schisandrin B-containing liposomes used as carrier of doxorubicin,the doxorubicin-loading efficiency and particle size distribution were determined using column chromatography.The acute toxicity and anti-tumor effect of the doxorubicin carrier with schisandrin B-containing lipsomes were evaluated in Kunming mice and animal S-180 tumor model.It was shown that the doxorubicin-loading efficiency was(96.45 ±1.86)%(n = 3),and the particle size distribution was(153.2±19.6) nm(n =3),and Zeta potential was(-38.6±2.5) mV.Schisandrin B-containing liposomes significantly reduced the toxicity of doxorubicin and improve its anti-tumor effect.
