摘要
采用浓度梯度筛选的方法,从铜污染土壤中分离筛选到一株耐受铜的菌株USTB-F,最高耐受浓度为560 m.gL-1.通过生理生化实验和16SrDNA序列分析鉴定属于不动杆菌属,命名为Acinetobactersp.USTB-F.用单因素方法得到USTB-F的最适生长条件为:T=30℃;pH=6.0;NaC l浓度为0.3%.对USTB-F富集机理做了初步研究,ICP-OES测定数据表明USTB-F菌对铜离子有富集功能,被吸收的铜离子有84.97%分布在细胞质内,其余主要分布在细胞膜和细胞壁上.
A strain capable of tolerating copper was isolated from heavy metal contaminated soils through concentration gradient screening technique,with the highest tolerating concentration of 560 mg·L-1.According to physiological and biochemical experiments and 16SrDNA sequence analysis,it was identified Acinetobacter sp..By using the single factor method,the optimum growth conditions were determined as follow: T=30 ℃,pH=6.0,NaCl concentration 0.3%.Measured and analyzed by ICP-OES,Cu2+ was accumulated by USTB-F.The distribution of adsorbed Cu2+ was 84.97% in cytoplasm,the rest mainly in cytolemma and cytoderm.
