摘要
目的:动态观察基质金属蛋白酶-9(matrix metallo proteinase-9,MMP-9)在阿霉素肾病大鼠肾小球内的表达,探讨其变化在肾小球疾病中的作用。方法:采用大鼠尾静脉单剂量注射阿霉素(浓度2mg/ml,剂量7.5mg/kg)建立阿霉素肾病模型,生化检测白蛋白变化,尿液检测24h尿蛋白定量,光镜下观察肾小球形态学改变,免疫组化检测肾组织MMP-9和纤维连接蛋白(FN)的表达。结果:阿霉素肾病组大鼠第4周时出现出现大量蛋白尿、低蛋白血症,与对照组相比,差异具有显著性(P<0.01);电镜下可见肾小球足突广泛融合;第8周部分肾小球发生局灶性硬化;MMP-9在肾小球足细胞的表达随着肾小球病变的加重先增加后减少;FN在肾小球内的表达随着肾小球病变的加重而逐渐增加。24h尿蛋白定量与MMP-9表达呈负相关,与FN表达成正相关,肾小球MMP-9与FN表达呈负相关。结论:阿霉素肾病组大鼠肾小球MMP-9和FN在不同阶段表达的变化可能参与了肾小球硬化的发生发展过程。
Objective:To investigate the expression of MMP -9 in glomerular in the rat model of adriamycin - induced nephropathy,and to explore the changes in the role of glomerular disease.Methods:The rats model of nephropathy was established by tail vein injection of adriamycin(concentration 2mg/ml,dose 7.5 mg/kg).The albumin changes were examined by biochemistry means,the 24 hour urine protein quota urine was examined in urine.The expressions of MMP -9 and fibronectin(FN) in the glomerular were observed by immunohistochemistry.Result:Cmpared with the control group,the adriamycin nephropathy group have lots of albuminuria,hypoalbuminemia,with significant difference(P0.01).A wide range of glomerular foot process fusion can be seen by electron microscope,part of the occurrence can be seen focal glomerular sclerosis at the eighth week。The expression MMP - 9 in glomerular podocyte is gradually increased in the first weeks and induced in the following weeks with the aggravated local lession in glomerular.But the expression of FN in glomerular is gradually increased.The 24 hour urinary protein was negatively correlated with expression of MMP -9,but positively correlated with FN.The expression of MMP -9 and FN in glomerular are signifi- cantly negativel correlated.Conclusion:that The expression of MMP - 9 and FN in different stages in Adriamycin - induced nephropathy rats glomerular may be involved in the process of the development of glomerular sclerosis.
