摘要
目的:观察大鼠弥漫性脑损伤(DBI)后不同时间皮质和海马中5-溴-2-脱氧尿苷(BrdU)和巢蛋白(Nes-tin)蛋白的变化,探讨其与脑损伤时间的关系。方法:成年健康SD大鼠36只,随机分为对照组(6只)和实验组(30只)。实验组大鼠制作DBI模型后用免疫组织化学方法观察脑损伤后不同时间(1、3、7、14和28d,n=6)皮质和海马中BrdU和Nestin蛋白的表达。结果:与对照组相比,实验组大鼠DBI后不同时间脑皮质、海马中的BrdU及Nes-tin阳性细胞均增加(BrdU:F皮质=141.447,F海马=48.361,P均<0.001;Nestin:F皮质=94.159,F海马=98.183,P均<0.001)。伤后1d,皮质和海马内就有BrdU和Nestin阳性细胞的大量表达;皮质内BrdU阳性细胞的表达于3d达到高峰,Nestin阳性细胞的表达于7d达到高峰,均持续至第14天后开始下降;海马内BrdU和Nestin阳性细胞的表达于第7天达到高峰,其后逐渐下降。结论:BrdU和Nestin蛋白表达所呈现出的时序性变化规律,可望用于DBI损伤时间的推断。
Aim:To observe the alteration of BrdU and Nestin positiveness cells in the diffuse brain injury(DBI) rat and its relation to the injury intervals.Methods:A total of 36 healthy adult SD rats were randomly divided into control and experimental groups.Based on Marmarou's model of DBI and took the samples at 1 d,3 d,7 d,14 d,and 28 d after brain injury.The expressions of BrdU and Nestin were observed by immunohistochemical staining in combination with imaging analysis.Results:Compared with the control group,at 1 day post-trauma,a significant increase in BrdU and Nestin postive cells was observed in the cortex and hippocampus tissue(BrdU:FCortex=141.447,FHippocampus=48.361,P0.001;Nestin:FCortex=94.159,FHippocampus=98.18,P0.001).The expression level of BrdU peaked at 3 days after injury and Nestin peaked at 7 days after injury in cortex tissue,then they decreased gradully at 14 days after injury.The expression level of BrdU and Nestin peaked at 7 days in hippocampus tissue,then they decreased gradually.Conclusion:Time-order expression of BrdU and Nestin could be used to estimate the time of brain injury in forensic pathology.
