不同强度电针对单纯性肥胖大鼠脂肪组织C-反应蛋白、白细胞介素6基因表达的影响 被引量:4

Effect of Different Intensities of Electroacupuncture on the Expression of C-reactive Protein and Interleukin-6 in Adipose Tissues of Simple Obesity Rats

摘要 目的观察电针对单纯性肥胖大鼠脂肪组织C-反应蛋白(CRP)、白细胞介素6(IL-6)表达的影响,并比较不同强度电针刺激的减肥效应差异。方法用普通鼠饲料喂养的SD雄性大鼠10只作为对照组;用高脂饲料喂养制作肥胖大鼠模型,并将造模成功的肥胖大鼠30只随机分为模型组、针刺1组、针刺2组,每组10只。针刺1组、针刺2组针刺一侧足三里和三阴交穴,并分别施以强度为1V和2V的电针刺激,每次治疗10min,每日1次,连续治疗14d。分别计算各组大鼠治疗前后的Lee’s指数,实验室检测大鼠血胆固醇(TC)、甘油三酯(TG)、高密度脂蛋白胆固醇(HDL-C)、低密度脂蛋白胆固醇(LDL-C),并采用RT-PCR技术测定脂肪组织CRP、IL-6基因mRNA表达水平。结果治疗后两个针刺组大鼠Lee’s指数、脂肪组织CRP、IL-6基因mRNA表达以及TC和TG含量与模型组比较均有明显下降(P<0.05或P<0.01),且针刺2组大鼠较针刺1组下降更为明显(P<0.05)。两个针刺组LDL-C、HDL-C的含量与模型组比较均有统计学意义(P<0.05或P<0.01),但针刺1组与针刺2组比较差异无统计学意义(P>0.05)。结论针刺能降低肥胖大鼠脂肪组织CRP、IL-6基因表达,改善肥胖大鼠机体的炎性反应状态,且2V电针刺激比1V电针刺激效果更好。 Objective To investigate the effect of electroacupuncture on the expression of C-reactive protein(CRP) and interleukin-6(IL-6) in adipose tissues of rats with simple obesity and compare the difference of different intensities of electroacupuncture for weight loss.Methods Ten male SD rats on normal diet were taken as the control group in the experiment.The obesity rat models were made by high fat diet and the 30 successful modles were randomized into model group,acupuncture group Ⅰ,and acupuncture group Ⅱ,with 10 in each.The acupuncture group Ⅰ and Ⅱ were treated with electroacupuncture at unilateral Zusanli(ST 36) and Sanyinjiao(SP 6),and stimulated by 1V and 2V respectively,10min each time,once a day,14 days in succession.Lee’s indices of all groups before and after treatment were calculated respectively,total cholesterol(TC),triglyceride(TG),high density lipoprotein cholesterol(HDL-C),and low density lipoprotein cholesterol(LDL-C) were determined,and the expression of CRP and IL-6 mRNA in adipose tissues were detected by RT-PCR technique.Results After electroacupuncture,Lee’s indices,expression of CRP and IL-6 mRNA in adipose tissues,the content of TC and TG of acupuncture groups were significantly decreased in comparing with the model group(P〈0.05 or P〈0.01),and the decrease in the acupuncture group Ⅱ was more significant than in the acupuncture group Ⅰ(P〈0.05).The content of LDL-C and HDL-C of acupuncture groups was statistically different from that of the model group(P〈0.05 or P〈0.01),but there was no statistical difference between acupuncture group Ⅰ and Ⅱ(P〉0.05).Conclusion Electroacupuncture decreases the expression of CRP and IL-6 mRNA in adipose tissues and improves the inflammatory state of obese rats,in addition,2V stimulation produces better effect than 1V stimulation.
