摘要 猕猴桃营养全面、丰富,含有10多种氨基酸、矿物质、胡萝卜素和多种维生素,对保持人体健康,防病治病具有重要的作用。维生素C又称L-抗坏血酸(ascorbic acid),是含有6碳的不饱和多羟化合物,在医药及营养保健方面的用途十分广泛。维生素C的分析方法有多种,但这些方法往往操作复杂费时,在样品处理过程中很容易使维生素C损失,且灵敏度和精确度不高;而高效液相色谱法(HPLC)具有较高的准确度、精密度和灵敏度,简便快速,更加适用于维生素C的测定。本研究采用高效液相色谱法(HPLC),探究猕猴桃中维生素C的最佳分析条件。通过大量的实验研究,表明以甲醇:纯水=2:8为流动相,流动相的速度为1.2ml/min,紫外检测波长为254nm,柱温在25℃为猕猴桃中维生素C的最佳HPLC分析条件。在此条件下分析猕猴桃中维生素C的含量、稳定性、回收率,所建立的标准曲线具有良好的线性关系,其相关系数R^2=0.9997;其平均回收率为100.21%,相对标准偏差小于1.47%;测得猕猴桃中维生素C的含量为107.33 mg/100g。所建立的方法简单、快速、准确度高,适宜猕猴桃中维生素C含量的分析研究。 The Chinese goosebeeiy's nutrition is comprehensive and abundant,containing more than 10 kinds of amino acids,minerals,carotene and vitamins.It plays an important role in maintaining human health,disease prevention and control.Vitamin C,also known as L-ascorbic acid(ascorbic acid),that contains 6 carbons unsaturated hydroxyl compounds,pharmaceutical and nutritional care in the use of wide-ranging.There are some methods for analyzing vitamin C,but these methods are often complex and time-consuming operation,the sample processing is very easy for the loss of vitamin C,and the sensitivity and accuracy is not high.However,the high performance liquid chromatography(HPLC) with high accuracy, precision and sensitivity,it is simple,rapid and more suitable for the determination of vitamin C.In this study,HPLC was performed to determine the content of vitamin C in the Chinese goosebeery.In this way,the content,the stability and the returns-ratio of the vitamin C in the Chinese goosebeery were analyzed under the condition of that the mixture of methanol mixed with pure water by 2:8 by volume was taken as flowing,and its speed was 1.2 ml/min,and the ultraviolet examination wave length was 254 ran with the column temperature under the 25'C.The good linear relations were gotten through the linear return with its correlation coefficient being R^2=0.9997.Its average returns-ratio was 100.21%.The relative standard deviation was smaller than1.47%.The content of vitamin C in the Chinese goosebeery was 107.33 mg/100 g.The method established was simple,fast and accurate,which is suitable to analyze the content of Vitamin C in the Chinese goosebeery.
