白黎芦醇对人口腔鳞癌KB细胞增殖和凋亡的影响及其机制 被引量:2

The Research of Anti-Proliferation and Induction of Apoptosis of Resveratrol on Human oral Squamous Cell Carcinoma KB Cells in Vitro

摘要 目的:探讨白藜芦醇对人口腔鳞癌KB细胞的增殖及凋亡影响。方法:应用MTT比色法检测Res对KB细胞体外生长的抑制作用;流式细胞仪测定细胞的周期和凋亡;Hoechst33258荧光染料染色,显示凋亡细胞形态;Western blot方法检测survivin、Caspase-3、Smac蛋白的表达。结果:Res能以剂量和时间依赖的方式抑制KB细胞生长,以剂量依赖的方式使KB细胞周期阻滞在S期、诱导细胞凋亡;Hoechst33258染色,可见细胞呈明显的凋亡特征;Western blot方法测得Res下调KB细胞的Survivin蛋白表达水平、增加KB细胞胞浆的Smac蛋白含量(P<0.01)、同时活化Caspase-3蛋白。结论:Res可抑制KB增殖,使细胞周期呈S阻滞并可诱导细胞发生凋亡,Survivin、Caspase-3和Smac基因参与了Res诱导KB细胞凋亡的作用。 Objective:To study the resveratrol on human oral squamous cell carcinoma cells proliferation and apoptosis.Methods:MTT colorimetric assay of KB cell in vitro growth inhibition;Cell cycle phase distribution and hypodiploid DNA was determined by flow cytometry;Hoechst33258 fluorescent dye staining,indicating apoptotic cell morphology;Western-blot method of detection of survivin,Caspase-3,Smac protein expression.Results:Res able to dose-and time-dependent manner inhibited KB cell growth;A dose dependent manner makes KB cell cycle arrest in S phase and induce apoptosis;Hoechst33258 staining,showing characteristics of apoptosis cells were significantly;Western-blot method measured Res down Survivin protein expression in KB cells,and increase the KB cell cytoplasm Smac protein content(P0.01),Caspase-3 protein was activated.Conclusion:Res inhibits proliferation of KB cell,the cell cycle arrest was S can induce apoptosis,Survivin,Caspase-3 and Smac gene involved in apoptosis of KB cell.
