摘要
目的:观察莪术黄芪超滤膜提取物对人SGC7901细胞是否具有凋亡诱导作用。方法:四氮唑蓝比色法(MTT法)观察莪术黄芪超滤膜提取物对人SGC7901细胞细胞的抑制率;应用流式细胞术,DNA凝胶电泳方法检测细胞凋亡的发生。结果:(1)莪术黄芪超滤膜提取物对人SGC7901细胞的生长有明显的抑制作用,与对照组相比有显著性差异(P<0.01),抑制作用与剂量及时间呈显著性正相关。(2)莪术、黄芪超滤膜提取物诱导人SGC7901细胞发生凋亡,1.6g/L处理,72h效果最佳。结论:莪术黄芪超滤膜提取物抑制人SGC7901细胞生长并促其凋亡。
Objective:To observe the effect of the ultra-filtration extract from Rhizoma Curcumae and Astragalus Membranaceous on inducing human SGC7901 cell apoptosis.Methods:SGC7901 cell proliferation was detected by MTT colorimetry.Apoptosis was observed by flow cytometry and agarose gel electrophoresis.Results:The ultra-filtration extract from Rhizoma Curcumae and Astragalus Membranaceous could significantly inhibit SGC7901 cell proliferation and there was a significant difference compared with that in the control groups(P0.01).There was a remarkable positive correlation between drug concentration of time and the inhibitory rates;The ultra-filtration extract from Rhizoma Curcumae and Astragalus Membranaceous could induce SGC7901 cell apoptosis.An optimal result is achieved with 1.6g/L for 72h.Conclusion:The ultra-filtration extract from Rhizoma Curcumae and Astragalus Memb-ranaceous could induce SGC7901 cell apoptosis.
