应激性生殖内分泌功能低下动物模型 被引量:15

ANIMAL MODELS OF STRESS REPRODUCTION ENDOCRINE DYSFUNCTION

摘要 目的 :观察应激对动物生殖内分泌的影响 ,建立符合临床需要的生殖内分泌紊乱的动物模型。方法 :用悬吊和制动方法造成动物急性或重复应激状态 ,用放射免疫结合的方法测定血浆有关性激素的水平 ,用阴道涂片的方法观察雌性动物的性周期 ,用常规方法观察雄性动物的性行为。结果 :急性制动应激和急性悬吊应激 1,2 ,3h均能使雄性小鼠血浆睾酮水平显著降低 ,皮质酮水平则明显升高。大鼠急性制动应激 3h ,可见血浆睾酮水平及睾丸中合成酶 17α 羟化酶、17,2 0 羟甾脱氢酶和 3 β 裂解酶活性显著降低 ,皮质酮水平则明显升高。重复悬吊应激可使雄性小鼠性行为低下或缺损 ,雌性小鼠性周期紊乱 ,分别可见血浆睾酮和雌二醇水平降低。结论 :应激可使生殖内分泌功能低下或紊乱 ,在此过程中性激素降低起着重要的作用。提示临床上各种躯体或心理应激引起的生殖内分泌功能低下或紊乱也与相关性激素降低或分泌节律失常有关 。 OBJECTIVE:To establish stress repioduction endocrine dysfunction(SRED) model in mice or rats for observation of the stress effects on function of reproduction endocrine system.METHODS:Acute and repeated stress conditions in mice or rats were induced by hanging and immobilizing methods and the plasma level of related steroid hormones were detected by RIA.Their sexual cycles or behaviors were detected by daily vaginal swabbing or conventional method.RESULTS:In male mice,the plasma levels of testosterone were significantly decreased and plasma levels of corticosterone were elevated after1,2,and 3 hours of hanging or immobilizing stress.The activities of testicular 17α hydrixylase,17,20 lyse and 3 β hydroxysteroid dehydrogenase in male rats were significantly reduced by 3 hours of acute immobilizing stress.The sexual behaviors of male mice and sexual cycles of femal mice were reduced and disturbed by repeated hanging stress of 12 consecuted days and their plasma levels of testosterone or estradiol were also reduced in the same time.CONCLUSION:The function of reproduction endicrine system of mice or rats might be reduced or disturbed by stress and sexual hormones played an important role in the course of time.Animal models of SRED established in the study might be useful in drug researches.
