摘要 目的 了解α-平滑肌肌动蛋白(α-SMA)在肾脏胚胎发育过程中的表达规律及α-SMA变化与肾脏成熟的关系。方法 用免疫组化链霉菌素亲生物素-过氧化物酶连接法(SP)测定不同胎龄的胚胎肾α-SMA的表达。结果 α-SMA在整个肾脏的发育过程中经历了早期阶段的无表达,较为成熟的肾小球系膜区呈强阳性,成熟肾小球呈阴性表达。结论 α-SMA在肾脏的表达具有一定的时空顺序,遵循一个离心方式,肾小球内α-SMA的表达是短暂的发育现象。 Objective To determine whether α smooth muscle actin (α SMA) is expressed in the developing kidney as well as the changes associated with maturation and to clarify its biological function. Methods Ten fetal and two normal adult kidneys were examined by immunocytochemistry (Strepavidin Peroxidase conjugated method, SP), using monoclonal mouse anti human antibody. Results Developing glomeruli show a gradient of expression of α SMA, ranging from absence in the early stages (vesicular, comma shap) of glomerulogensis to pronounced mesangial expression in more mature glomeruli However it became negative in the mature glomeruli. Conclusion The expression of α SMA in developing kidney has chronological and spatial sequence. Glomerular mesangial expression of α SMA is a transient phenomenon associated with active glomerulogenesis. Once glomerular development is completed, α SMA will disappear completely from glomeruli.
