摘要
目的:探讨同基原赤、白芍增强小鼠免疫作用的异同并比较其化学成分的相似度,寻求免疫作用的物质基础。方法:腹腔注射环磷酰胺复制免疫低下动物模型,随机分为赤、白芍高、中、低剂量组、空白组、阳性对照组,测定碳粒廓清、迟发型超敏反应、血清溶血素含量以及巨噬细胞吞噬功能;采用HPLC法分析比较起效赤、白芍提取物中的化学成分。结果:赤、白芍给药组与对照组相比,均能显著提高免疫低下模型小鼠免疫器官质量(P<0.05)、极显著增强巨噬细胞吞噬能力(P<0.01)、极显著升高血清溶血素的含量(P<0.01)。此外,HPLC图谱显示本次试验中的赤、白芍起效成分提取物在230nm下具有17个共有峰,相似性为81.29%。结论:同基原赤、白芍化学成分具有相似性,在本次试验中增强小鼠免疫的作用相似,提示其相似的化学成分可能为增强免疫的物质基础。
OBJECTIVE To study the effects of Radix Paeoniae Alba (PRA) and Radix Paeoniae Rubra (PRR) with some origin on immunological function of ICR mice, and to investigate the material foundation of pharmacological effects by comparing the chemical constituents. METItODS Cyclophosphamide (CTX, ip) induced immunosuppressed mice were randomly divided into 9 groups. Tested groups intaked 50 -500 mg·mL-1 PRA or 70 - 700 mg.mL-1PRR, the group intaked 1mg.mL-1 thymosin as positive control. Carbon granule clearance, delayed type hypersensitivity, the contents of hemonlysin and phagocytic activity of peritoneal macrop hages were measured. And then compared PRA and PRR extract by HPLC. RESULTS PRA and PRR herb extract could obviously enhance the weight of immune organ (P〈0. 05), the percentage of phagocytosis, the macrophage phagocytosis index (P〈0. 01) as well as the hemolysin antibody level (P〈0. 01) of treated groups. The results of HPLC results indicated 17 common characteristic peaks (similarity is 81.29%) in PRA and PRR extract under 230 nm. CONCLUSION In this research, the same origin PRA and PRR showed a similar immuno-enhancing capacity in mice, the similar inclusions are respond for the immuno-enhancing activity.
