摘要
目的:了解0139型霍乱弧菌在四种4号琼脂平板上的生长特性,为选择其合适的培养基提供参考。方法:将本实验室分离的0139型霍乱菌株经增菌后按一定比例稀释,分别接种到正常人群肛拭样中,按照《霍乱防治手册》(第五版)同时用四种4号琼脂进行分离培养。结果:四种4号琼脂平板均发现有可疑菌落O139型霍乱弧菌生长。在"北京陆桥"、"杭州天和"平板上10 h^12 h内能分离培养出可供试验的可疑菌落;16 h分离培养后菌落特征分别为:在"杭州天和"平板上呈圆形、中心深褐色、边缘无色、2 mm^3 mm大小菌落,在"北京陆桥"平板上呈圆形、部分菌落边缘有1~2个棘、似椭圆、中心褐色凹陷呈脐窝状、周围乳白色、2 mm^3 mm大小菌落,在"广东环凯"平板上呈圆形、中心浅褐色呈点状隆起、周围无色透明、2 mm大小菌落,在"青岛海博"平板上呈圆形、乳白色、扁平、1 mm大小菌落。结论:四种4号琼脂培养基均能分离出O139型霍乱弧菌;而菌落大小、形状、色泽等菌落特征差异明显,部分4号平板上其典型菌落特征未见文献报道;同时在有的平板上生长较快,能及时检测病原菌。此次实验提示,"杭州天和"等厂家生产的4号琼脂比较适合本地区O139型霍乱弧菌分离培养。
Objective:Understanding the growth characteristics of Vibrio cholerae O139 in four kinds of NO.4 agar plates,and providing a reference for the selection of the suitable medium.Methods:The Cholera O139 stains,which isolated in our laboratory,was diluted by a certain percentage after enrichment,then inoculated into the anal swab samples of the normal population,isolated and cultured using agar plate,in accordance with "cholera prevention manual"(fifth edition).Results: The suspicious colony of Vibrio cholerae O139 was growing on four kinds of NO.4 plates.The suspicious colony that available for testing can be isolated in the plates which produced by "Beijing Luqiao","Hangzhou Tianhe" within 10-12h.The characteristics of isolated colonies after 16h are as follows: The colony in the plate which produced by "Hangzhou Tianhe",was 2-3mm and round,dark brown in the center,colorless in the edge.The colony in the plate which produced by "Beijing Luqiao",was 2-3mm and round,brown in the center and hollowed as umbilical fossa,milky white in the surrounding,and part of colony had 1-2 spines in the edge,like oval.The colony in the plate which produced by "Guangzhou huankai ",was 2mm and round,light brown in the center,spotty uplift,colorless and transparent in the surrounding.The colony in the plate which produced by "Qindao Haibo ",was 1mm and round,milky white and flat.Conclusion: The Vibrio cholerae O139 can be isolated in four kinds of NO.4 plates,but it's colony characteristics such as colony size,shape,color was a significant difference,and part of the typical characteristics wasn't been reported.And the colony grew faster in some kinds of plates,and can be detected in time.The experiment suggested that,the NO.4 agar plates produced by "Hangzhou Tianhe " and other manufacturers are more suitable for isolation of Vibrio cholerae O139 in this region.
