摘要
目的 探讨TPA对人喉鳞癌细胞的抗增殖作用。方法 应用MTT、流式细胞 (FCM)技术对Hep - 2细胞进行观察和检测。结果 10nMTPA作用 12h ,可明显抑制Hep - 2细胞的增殖 ;下调PCNA的表达量 ;细胞分裂阻滞于G1期。结论 TPA能够抑制人喉鳞癌细胞的增殖 ,这种作用与细胞核PCNA的变化有密切的关系。
Objective Antiproliferative effects of TPA on human laryngeal squamous cancinoma cells (Hep 2) was studied.Methods MTT was performed.Cell cycles and expression of PCNA were detected by FCM.Results When Hep 2 cells were treated with 10 nM TPA for 12 hours,cell proliferation was obviously inhibited;expression of PCNA in cell nuclei was deregulated,and cell cycle was arrested in the G 1 phase after TPA treatment.Conclusion TPA can inhibit proliferation of human laryngeal squamous carcinoma cells,this effect is associated with changes of PCNA expression in cell nuclei.
