反相高效液相色谱法测定硫酸长春新碱的含量

Determination of Vincristine in Tumor by RP-HPLC

目的 :本文采用外标法测定了小鼠肿瘤组织内的长春新碱的含量。方法 :应用0 04mol/L磷酸缓冲液 -甲醇 (45∶55 ,pH3)为流动相 ,在C185μm色谱柱上进行测定。结果 :长春新碱的检测线性范围为0 01875～0 6μg/ml,γ=0 9998。回收率为89 6 %(n=10) ,RSD为0 86 %。结论 :本法简便、快速、灵敏。

OBJECTIVE: This paper reports a RP-HPLC method for the determination of vincristine in mouse tumor tissue METHODS:Vincristine was separated and determined on a reverse-phase C18 5μm column with a mobile phase of 0 04mol/L phosphate buffer solution-MeOH(45:55,pH3), detected at 254nm and no internal standard RESULTS: The calibration curve was linear(γ=0 9 998) within the range of 0 01 875～0 6μg/ml for vincristine The recovery ratio of vincristine was 89 6%(n=10) The relative standard deviation(RSD) was 0 86% CONCLUSION:This method is simple and accurate
