摘要
目的:建立金益排石颗粒中大黄酚含量的微乳液相色谱测定法。方法:以微乳2.5%(w/v)十二烷基硫酸钠(SDS)-0.1%(v/v)正辛烷-10.0%(v/v)正丁醇-0.5%三乙胺(v/v)(磷酸调pH至3.0)为流动相,色谱柱采用Hypersil ODS_2色谱柱(250 mm×4.6 mm,5μm),柱温28℃,流速1 ml·min^(-1),检测波长254 nm。结果:大黄酚在3.9~39.0μg·ml^(-1)范围内,与峰面积呈良好的线性关系(r=0.999 8),平均加样回收率为99.66%,RSD为0.64%(n=6)。结论:本法简便、准确,可用于金益排石颗粒中大黄酚的含量测定。
Objective : To develop a method for determination of chrysophanol in Jiyi Paishi granules by microemulsion liquid chro- matography. Method: The mobile phase was 2.5 % (w/v) sodium dodecyl sulfate-0. 1% (v/v)n-octane-10. 0 % (v/v) n-butanol-0. 5 % tfiethylamine (v/v),adjusting pH to 3.0 with phosphoric acid. The analytical column was Hypcrsil ODS2 (250 mmx 4. 6 mm,5 ~Lm) with the column temperature of 28 ~C ,and the flow rate was 1 ml.min 1 and detection wavelength was 254 nm. Result: The cali- bration curve was linear within the rang of 3.9-39. 0μg· ml^ - 1 for chrysophanol with r = 0. 999 8, the average sample-adding recovery was 99.66% with RSD of 0. 64% ( n = 6 ). Conclusion : The method is simple and accurate, and can be used for the quality control of Jiyi Palshi granules.
